Firefly luciferase facilitates the conversion of luciferin to oxyluciferin in conditions where ATP, magnesium ions, and oxygen are present, resulting in the emission of bioluminescence at a wavelength of about 560 nm, detectable with a luminometer. CleanRNA™ Firefly Luciferase-mRNA (low dsRNA) is an mRNA that has undergone 5' end capping, 3' end polyadenylation, and modification with N1-Methyl-Pseudouridine Triphosphate (N1-Me-Pseudo UTP). Upon transfection into cells, it allows for the intracellular expression of the firefly luciferase protein. The product is generated via in vitro transcription with an optimized wild-type T7 RNA polymerase, which minimizes dsRNA content in the mRNA solution, thereby enhancing the quality of the Firefly Luciferase-mRNA. Additionally, the product incorporates 3-O-Methyl-Guanine (3-O-Me-GAG) for cotranscriptional capping of the mRNA, creating a Cap 1 structure that boosts the stability and translational efficiency of the Firefly Luciferase-mRNA.

The products are shipped with dry ice and can be stored at -80℃ for two year.

 

Notes

1. Upon arrival, immediately store the samples at -80°C and avoid repeated freeze-thaw.

2. For the first use, be sure to wear disposable gloves, dissolve the samples on ice, and use RNase-free reagents and consumables to avoid contamination.

3. After the initial dissolution of the samples, they can be gently centrifuged and then aliquoted, avoiding repeated freezing and thawing.
